It seems that he has lost them somewhere around town and has asked you to help him track them down. Where to find the Mayor’s Shorts in Stardew Valley

You can find the Mayor’s shorts in Marine’s bedroom in Stardew Valley, but you won’t be able to go there immediately as you need to increase your friendship with Marine to two hearts beforehand. You can do this by completing quests for her or by giving her gifts. If you go with the gift option, you can’t just give her anything, as there are specific types of gifts certain NPCs like and do not like. Players should also keep in mind that you will only have two chances to give an NPC a gift each week, so you better make it a good one. Once you find a gift for Marine, you can typically find her at her farm, which is south of yours. Once you find her, you can give her one of the gifts and then back the next day and give her another. Unfortunately, after that you’ll have to wait an in-game week to give her another. Repeat this process until eventually, you reach two friendship hearts and you’ll be able to go into her room.

After going upstairs, locate her room, and the shorts will be at the end of her bed. Now all you need to do is take the shorts and give them back to Mayor Lewis, who usually be found in the Mayor’s Manor. After completing the Stardew Valley Mayor’s Shorts quest, make sure to go into your journal to accept the reward which is 750g.
